Datopotamab deruxtecan (Dato-DXd) is approved for the second-line treatment of EGFR-mutated non-small cell lung cancer (NSCLC) and has also demonstrated significant clinical activity in breast cancer. The open-label, randomized, phase 3 TROPION-Breast02 trial evaluated the efficacy and safety of Dato-DXd compared with chemotherapy in patients with unresectable or metastatic breast cancer who were ineligible for PD-1/PD-L1 inhibitor therapy. Findings from the study supported the approval of Dato-DXd for this patient population and informed updates to the NCCN Clinical Practice Guidelines in Oncology for breast cancer.
